What it is

Your body's main antioxidant. Cells make it from cysteine, glycine, and glutamate. Direct oral supplementation has been controversial due to poor absorption.

Why people take it

Senaste liposomal forms and Setria® show meaningful blood-level increases. NAC + glycine remains cheaper and works upstream.

Our verdict

Take NAC + glycine (GlyNAC) unless you specifically want the direct form for a targeted use.

Structure & class

γ-Glutamyl-cysteinyl-glycine tripeptide.

Mechanism & clinical data

Richie 2015 showed 250 mg Setria® raised GSH stores. Liposomal forms improve on standard oral.

Pharmacokinetics

Rapid.

Where this dose comes from

The trial dose above is our editorial pick from the published human trials for this ingredient. We choose the dose that the best-available studies used on the endpoint most relevant to longevity. The range shows the lowest and highest doses tested in published trials. The landmark studies below are the papers we read to set it. You can click through and check our work.

When new trials publish, our automated sweeps flag them. If a new study materially changes the dose, we update it through editorial review and record the old dose, new dose, and the triggering study in every affected product's recommendation history. The dose is never changed automatically.

EU Court sharpens the bar for botanical health claims
A Court of Justice of the European Union ruling in april 2025 further tightened the requirements for how botanical ingredients (herbs, plant extracts) can be marketed with health claims in the EU, effectively removing much of the flexibility of the on-hold list.
  • 2024-06 Regelverk confirmed
EU report identifies 117 supplement substances as potential health risks
The HoA Working Group on Food Supplements (26 European countries) published its first report evaluating 117 substances used in food supplements. Of these, 65 were assessed as likely novel foods, 6 were already regulated, 34 had insufficient data, and 12 were prioritized for Article 8 restriction under Regulation (EC) No 1925/2006. The 12 priority substances include ashwagandha, curcumin, melatonin, piperine, St. John's wort, coumarin, p-synephrine, maca, holy basil, Melaleuca, black cohosh, and L-tryptophan. The report drew on 1,500 RASFF alerts reviewed between 2017 and mid-2022.

    Richie et al. 2015
250 mg Setria®/dag raised body GSH stores over 6 months.
  • 2006-12 Regelverk confirmed
    EU Health Claims Regulation adopted
Regulation (EC) No 1924/2006 laid down harmonised rules for nutrition and health claims made on foods, including supplements. Only claims listed in the EU Register of authorised claims can be used. Rejected botanical claims are held in a separate on-hold list.

EU introduces the Novel Food Regulation
Regulation (EC) No 258/97 required any food or food ingredient without a history of consumption in the EU before 15 maj 1997 to be authorised before being placed on the market. This is the framework that captures NMN, spermidine as a supplement ingredient, urolithin A, and many other longevity actives.
2026 evidence refresh

Oral glutathione reliably raises body glutathione stores in humans, but the trial evidence for skin whitening and other marketed benefits is small and mixed.

In diabetic patients over 55 on top of standard anti-diabetic treatment, 500 mg/day oral glutathione significantly lowered HbA1c and oxidative DNA damage (8-OHdG) versus anti-diabetic treatment alone, but showed no extra benefit in the full adult diabetic cohort. source

Best-supported claim

Oral glutathione raises measurable glutathione levels in blood, erythrocytes, plasma, and buccal cells. Richie et al.'s 2014 randomized, double-blinded, placebokontrollerad trial in 54 healthy non-smoking adults found 6 months of 1,000 mg/day increased erythrocyte and plasma glutathione by 30-35% and buccal cell glutathione by 260%, with levels returning to baseline after a 1-month washout (https://pubmed.ncbi.nlm.nih.gov/24791752/). In elderly type 2 diabetics, Kalamkar et al.'s 2022 trial in 250 diabetic patients found 500 mg/day for 6 months reduced oxidative DNA damage (8-OHdG) and, in the over-55 subgroup, significantly lowered HbA1c (https://pmc.ncbi.nlm.nih.gov/articles/PMC9137531/).

Not supported by the evidence base

Glutathione is marketed heavily as an oral or IV skin-whitening agent and a general anti-aging antioxidant, but the skin-lightening evidence is thin and inconsistent. A review of three RCTs found one trial showed significant melanin reduction only in sun-exposed areas, one found glutathione performed no better than placebo, and a third found benefits limited to subjects over 40 in specific skin areas, with color improvements disappearing within 6 months of stopping (https://pmc.ncbi.nlm.nih.gov/articles/PMC7196133/). Marketers also imply oral glutathione directly patches cellular antioxidant defenses body-wide, but raising blood glutathione does not equal proven clinical benefit for aging, energy, or immunity outside the diabetic HbA1c signal seen in one trial (https://pubmed.ncbi.nlm.nih.gov/24791752/).

Typical dose

250-1000 mg/day oral reduced glutathione, most study doses landing at 500 mg/day

Säkerhet

Oral glutathione at doses up to 1,000 mg/day for 6 months has shown no significant adverse events versus placebo in trials (https://pubmed.ncbi.nlm.nih.gov/24791752/). Intravenous and injectable glutathione, common in skin-whitening clinics, is not well studied for long-term safety and is not the form used in most oral-supplementation RCTs. Personer with asthma have reported bronchospasm after inhaled or nebulized glutathione, a different route not relevant to oral capsules. Pregnant or breastfeeding people and those with sulfa allergies should check with a physician before use, since glutathione synthesis depends on sulfur amino acid metabolism (https://pmc.ncbi.nlm.nih.gov/articles/PMC7196133/).

Interactions worth flagging

  • maj theoretically alter chemotherapy drug efficacy by countering oxidative mechanisms some cancer therapies rely on
  • No well-established prescription drug interactions documented in oral RCTs to date
