Best-supported claim
NMN reliably raises blood NAD+ levels in humans. A 2024 metaanalys of 12 RCTs (513 participants) by Jiaqi Zhang and colleagues confirmed a significant rise in blood NAD after supplementation (https://pubmed.ncbi.nlm.nih.gov/39116016/). A separate 2024 metaanalys of 9 studies (412 participants) by Jian-Peng Wang and colleagues found NMN improved gait speed (SMD 0.34 m/s, p=0.033) and lowered ALT in middle-aged and elderly adults (https://pubmed.ncbi.nlm.nih.gov/39185644/). These are the two strongest signals in the human data.
Not supported by the evidence base
Marketers describe NMN as a reversal agent for aging, promising restored energy, sharper cognition, and extended lifespan. Human RCTs do not support this. Zhang's 2024 metaanalys found most clinically relevant outcomes, including fasting glucose, triglycerides, and cholesterol, showed no significant difference from placebo (https://pubmed.ncbi.nlm.nih.gov/39116016/). A 2025 review by Aisyah Kuerec and colleagues on muscle mass and function in adults over 60 found NMN and NR gave no significant improvement in skeletal muscle index, grip strength, knee extension, or short physical performance battery scores (https://pmc.ncbi.nlm.nih.gov/articles/PMC12022230/). No human trial has measured lifespan extension.
