What it is
Another route to raise NAD+, the same energy/repair molecule NMN targets. NR is one step further away from NAD+ than NMN, but it's the only one of the two that's been formally approved in the EU as safe.
Why people take it
NR raises NAD+ reliably in humans. Multiple 8 to 12 week studies show modest improvements in some biomarkers. Whether it makes you live longer is unproven, but for staying at status quo it has more safety data than almost any longevity ingredient.

The safer, more boring cousin to NMN. If you want a NAD+ booster and care about approvals, NR is the pragmatic choice.

Best-supported claim
NR consistently and reliably raises blood NAD+ levels, typically 2 to 3-fold, across nearly every human trial regardless of population or dose. Chao-Yi Wu and Edmarie Guzman-Velez's 2025 trial in long-COVID patients confirmed this pharmacodynamic effect using 2000 mg/day (https://pubmed.ncbi.nlm.nih.gov/41357333/). Yasir Elhassan and colleagues showed in 2019 that NR also reaches skeletal muscle in aged men, elevating the NAD+ metabolome and lowering circulating inflammatory cytokines after 21 days at 1000 mg/day (https://pubmed.ncbi.nlm.nih.gov/31412242/). This biomarker effect is the most reproducible finding in the NR literature.
Not supported by the evidence base
Marketers sell NR as an anti-aging, cognition-boosting, blood-pressure-lowering supplement. Human trials tell a different story on downstream effects. Miranda Orr's 2023 randomized trial in older adults with mild cognitive impairment found NR raised NAD+ 2.6-fold but produced no improvement on the Montreal Cognitive Assessment or other cognitive tests over 10 weeks (https://pubmed.ncbi.nlm.nih.gov/37994989/). The 2025 long-COVID trial found no significant benefit on cognition, fatigue, sleep, or mood despite the same NAD+ boost (https://pubmed.ncbi.nlm.nih.gov/41357333/). A 2025 pilot trial combining NR with exercise in hypertensive older adults found NR plus exercise was not superior to exercise alone for lowering systolic blood pressure (https://pubmed.ncbi.nlm.nih.gov/40770531/). Raising a biomarker is not the same as changing an outcome that matters.

The NR-SAFE trial in Parkinson's disease patients found high-dose NR safe but flagged it can accumulate certain metabolites that warrant monitoring in some populations. Long-term safety data beyond 6 months in healthy adults is limited.
