Spermine is synthesized from spermidine via spermine synthase, and is itself a substrate for further catabolism; it participates in chromatin stabilization, membrane stabilization, and has been implicated in mitochondrial function and cellular stress resistance.
Mechanism & clinical data
The foundational paper (Eisenberg et al. 2009, Nat Cell Biol) established that supplementation with spermidine (the polyamine one step upstream of spermine) extends lifespan in yeast, flies, worms, and enhances autophagy in human immune cells, with intracellular polyamine concentration (including spermine) declining during human aging. A human pharmacokinetic study (2023, PMC10143675) found that oral spermidine supplementation (15 mg/day for 5 days) significantly increased plasma spermine levels without affecting spermidine or putrescine levels, suggesting that dietary spermidine is presystemically converted to spermine, which is proposed as a mediator of at least part of spermidine's systemic effects. No dedicated RCT of oral spermine supplementation (as opposed to spermidine) was found in this search.
Pharmacokinetics
Not established for direct oral spermine supplementation in humans; plasma spermine elevation has been observed as a downstream consequence of oral spermidine dosing (15 mg/day) rather than direct spermine administration.
