NICE-PAD II Trial: Nicotinamide Riboside for Peripheral Artery Disease (Phase 3)

NICE-PAD II (NCT07782671) is a Phase 3 randomized trial testing nicotinamide riboside (NR) in people with lower-extremity peripheral artery disease. The trial was registered on ClinicalTrials.gov with a last-update date of 2026-08-24 and a status of not yet recruiting. It is a follow-up to the original NICE trial (NCT03743636), published in Nature Communications in 2024 (PMID 38871717), which tested 6 months of NR with and without resveratrol in 90 people with PAD. The original trial measured 6-minute walk distance at 6-month follow-up. NICE-PAD II is sponsored by Mary M. McDermott, MD, at Northwestern University. No enrollment target, primary outcome data, or results have been posted yet. The trial registration confirms continued academic interest in NR for vascular function but provides no new efficacy or safety evidence at this stage.
